Authorized Use of ImmuneOnco’s Jurkat-CSR Cell Line

         Recently ImmuneOnco has authorized the use of the independent intellectual property rights -- Jurkat-CSR cell line -- toseveral domestic partners (Sumgenbio, Mab-works, etc.). The cell line expresses CSR molecule, which is composed of the outer membrane of SIRP alpha, the hinge region of CD8 alpha, the transmembrane and intracellular region of CD28, and the signal transduction region of CD3z. Moreover, using CRISPR-Cas9 technology, we knocked out the hCD47 gene of Jurkat cell line.

This cell can specifically bind to hCD47 protein. After binding, it can initiate signal transduction and induce cell death through the binding of FasL (CD95L) and Fas (CD95) between cells. Activation-induced cell death (AICD) is initiated to induce cell death. Adding CD47 or SIRPa antibody can inhibit the activation of Jurkat-CSR cells by CD47 and prevent cell death. The chimeric protein cell line can be used to detect the cell biological activity of CD47/SIRP alpha blocker and to screen CD47 or SIRP alpha specific molecular drugs (monoclonal antibodies, gene recombinant proteins, targeted small molecular drugs, etc.).

In order to study the ability of CD47 target drugs to promote the detection of phagocytosis of cancer cells, traditional in vitro activity analysis methods for CD47 target drugs need to isolate lymphocytes from human peripheral blood by using lymphocyte separating fluid, further obtain monocytes by magnetic bead sorting method, and then differentiate them into macrophages through long-term stimulation and culture. This method has low operability, long cycle, high cost, low throughput and is difficult to be used for drug release detection. The Jurkat-CSR cell line can be used for screening, functional analysis and release detection of CD47 or SIRP alpha target drugs, and have the advantages of simple operation, short cycle, low cost, good repetition etc.
